RESUMEN
Los Péptidos Antimicrobianos (PAMs) son componentes indispensables de los mecanismos de defensa innatos de los seres vivos y actualmente son una alternativa para el combate de cepas bacterianas y fúngicas resistentes a los antimicrobianos convencionales. La producción de PAMs por vía recombinante es una opción viable para su obtención de forma masiva. En esta investigación se evaluó la expresión de algunos PAMs en las microalgas Chlamydomonas reinhardtii y Chlorella vulgaris y en la bacteria Escherichia coli. Los péptidos sujetos a estudio fueron la bacteriocina E-760 que ha demostrado tener actividad inhibitoria contra bacterias Gram negativas y Gram positivas, los PAMs de M. oleifera Mo-CBP3 (capacidad antifúngica) y FLO (efecto bactericida). El gen sintético E-760S fue insertado en el genoma nuclear de C. reinhardtii utilizando Agrobacterium tumefaciens. El rendimiento en la producción de E-760 recombinante fue de 0.14 a 0.36% del total de proteínas totales solubles (PTS) y mostró actividad inhibitoria moderada contra Staphylococcus aureus, Staphylococcus agalactiae, Enterococcus faecium, Pseudomonas aeruginosa y Klebsiella pneumoniae. El gen sintético FLOGS fue insertado en el genoma plastidial de C. vulgaris por medio de biobalística. El rendimiento en la producción de FLOGS fue de 0.066 a 0.09% del total de PTS, no mostró ningún efecto inhibitorio contra las cepas bacterianas evaluadas. Se obtuvieron las construcciones con los péptidos Mo-CBP3 y FLO en el vector pET32, así como la secuencia sintética denominada gBlock (Mo-CBP3 + FLO) clonada en el vector pET32. Estos resultados demuestran que las microalgas C. reinhardtii y C. vulgaris funcionan como plataformas estables para la producción de proteínas terapéuticas importantes.

Antimicrobial peptides (PAMs) are indispensable components of the innate defense mechanisms of living beings and are currently an alternative for combating bacterial and fungal strains resistant to conventional antimicrobials. The production of PAMs by recombinant way is a viable option to obtain them massively. In this research, the expression of some PAMs in the microalgae Chlamydomonas reinhardtii and Chlorella vulgaris and in the bacterium Escherichia coli were evaluated. The peptides under study were the bacteriocin E-760 that has been shown to have inhibitory activity against Gram-negative and Gram-positive bacteria, the PAMs of Moringa oleifera Mo-CBP3 (antifungal capacity) and FLO (bactericidal effect). The synthetic gene E-760S was inserted into the nuclear genome of C. reinhardtii using Agrobacterium tumefaciens. The yield in the production of recombinant E-760 was 0.14 to 0.36% of total soluble protein (TSP) and showed moderate inhibitory activity against Staphylococcus aureus, Staphylococcus agalactiae, Enterococcus faecium, Pseudomonas aeruginosa and Klebsiella pneumoniae. The synthetic gene FLOGS was inserted into the plastidial genome of C. vulgaris by biolistic via. The yield in the production of FLOGS was 0.066 to 0.09% of the total of TSP, did not show any inhibitory effect against the bacterial strains evaluated. The constructs were obtained with the peptides Mo-CBP3 and FLO in the vector pET32, as well as the synthetic sequence called gBlock (Mo-CBP3 + FLO) cloned in the vector pET32. These results demonstrate that microalgae C. reinhardtii and C. vulgaris function as stable platforms for the production of important therapeutic proteins.
