Resumen
Se conoce como huitlacoche a las agallas (tumores) comestibles inducidas por el hongo basidiomiceto Ustilago maydis DC Corda en las mazorcas de maíz (Zea mays L.). El huitlacoche tiene muchos compuestos fenólicos asociados con la capacidad de disminuir el riesgo de contraer enfermedades degenerativas como la diabetes. Este trastorno metabólico se caracteriza principalmente por hiperglicemia y se relaciona con el desarrollo de complicaciones que señalan al estrés oxidativo (EO) como la vía común que une los diferentes mecanismos de las patogénesis asociadas.
El objetivo de este trabajo fue estudiar el efecto del huitlacoche (Ustilago maydis-Zea mays) sobre algunos indicadores bioquímicos de estrés oxidativo en ratas diabéticas y normales. Para ello se utilizaron ratas Wistar machos de 250 - 300 g y se dividieron en tres subgrupos: ratas control alimentadas con croquetas de dieta base, ratas alimentadas con croquetas de dieta base adicionadas con huitlacoche crudo al 10% (base seca), y ratas alimentadas con croquetas de dieta base adicionadas con huitlacoche cocido (guisado) al 10% (base seca). La diabetes fue inducida con estreptozotocina (50 mg/kg). Las ratas recibieron su dieta respectiva durante 4 semanas y se monitorearon cada 7 días midiendo las concentraciones plasmáticas de trifosfato de adenosina (ATP), malondialdehído (MDA) y creatinina. Antes del sacrificio (día 28), se midieron el flujo urinario (FU), las concentraciones de glucosa y sodio y la masa excretada de proteínas (MEP). Además, con la finalidad de complementar la evaluación de la eficacia del tratamiento con huitlacoche sobre la diabetes y valorar el daño funcional de hígado y riñones, se realizaron pruebas adicionales a los 28 días del estudio, es decir, el día del sacrificio. Estas pruebas incluyeron cuantificaciones plasmáticas de las transaminasas glutámico pirúvica (TGP) y glutámico oxáloacética (TGO), así como de glucosa, ácido úrico, urea y nitrógeno ureico (BUN).
Los resultados obtenidos muestran que el FU y la MEP disminuyeron significativamente en ratas diabéticas respecto a los controles; el FU se normalizó en las diabéticas alimentadas con huitlacoche. Los niveles de glucosa disminuyeron notablemente en la orina y plasma de las ratas diabéticas alimentadas con huitlacoche, respecto a las alimentadas con dieta base. En el plasma de las ratas diabéticas alimentadas con huitlacoche, las concentraciones de ATP aumentaron y los niveles de MDA disminuyeron, respecto a las ratas alimentadas con dieta base. Las concentraciones plasmáticas de TGO y TGP disminuyeron considerablemente en ratas diabéticas cuyas dietas fueron suplementadas con huitlacoche, al igual que los niveles de glucosa y ácido úrico. Los niveles de urea y BUN disminuyeron significativamente en los grupos de ratas diabéticas alimentadas con huitlacoche con respecto a las ratas controles.
En base a los resultados obtenidos se propone el consumo de una dieta suplementada con huitlacoche para disminuir algunos indicadores de estrés oxidativo en individuos diabéticos, así como para disminuir las concentraciones de indicadores del daño funcional en hígado y riñones. Se concluye que el tratamiento térmico del huitlacoche no afecta sus efectos fisiológicos benéficos sobre dichos indicadores.
Summary
Edible galls (tumors) induced by the basidiomycete fungus Ustilago maydis DC Corda on maize (Zea mays L.) ears are known as huitlacoche. Huitlacoche contains many phenolic compounds associated with the capacity to diminish the risk of contracting degenerative diseases such as diabetes. This metabolic disorder is characterized by hyperglycemia and is related to the development of complications which point at oxidative stress as the common pathway from which the different mechanisms of associated pathogeneses diverge.
The objective of this work was to study the effect of huitlacoche (Ustilago maydis-Zea mays) on several biochemical indicators of oxidative stress in diabetic and normal rats. Wistar rats (250-300 g) were allotted into three subgroups: control rats fed with basal diet pellets, rats fed with basal diet pellets added with 10% (dry basis) raw huitlacoche, and rats fed with basal diet pellets added with 10% (dry basis) cooked (fried) huitlacoche. Diabetes was induced with streptozotocin (50 mg/kg). Rats were fed during 4 weeks and were monitored every 7 days by measuring plasma concentrations of adenosine triphosphate (ATP), malondialdehyde (MDA), and creatinine. Before sacrifice (on day 28), urinary flow (UF), and the concentrations of glucose, sodium and excreted protein mass (EPM) were measured. Also, in order to complement the evaluation of the efficacy of the treatment with huitlacoche on diabetes, and to evaluate the functional damage on liver and kidneys, additional test were carried out on day 28. These tests included measurements of plasma levels of glutamic-pyruvic transaminase (GPT) and glutamic-oxaloacetic transaminase (GOT), as well as glucose, uric acid, urea and ureic nitrogen (BUN).
Results show that UF and EPM were diminished significantly in diabetic rats with respect to the controls; UF was normalized in diabetic rats fed with huitlacoche. Glucose levels decreased markedly in urine and plasma from diabetic rats fed with huitlacoche, with respect to those fed with basal diet. ATP concentrations increased and MDA levels decreased in plasma from diabetic rats fed with huitlacoche, in relation to those fed with basal diet. Plasma levels of GPT and GOT were considerably diminished in diabetic rats fed with the huitlacoche amended diets, as well as glucose and uric acid levels. Urea and BUN levels decreased significantly in the diabetic rats fed with huitlacoche, with respect to the control rats.
Based on these results, the consumption of a diet supplemented with huitlacoche is proposed, in order to diminish several indicators of oxidative stress in diabetic individuals, as well as to diminish the concentrations of indicators of functional damage in liver and kidneys. It is concluded that the thermal treatment of huitlacoche does not affect its beneficial physiological effects on the above-mentioned indicators.
